Fixation in cytochemistry and electron-microscopy.

چکیده

Whenever it is our intention to study objects that are visible with the light-microscope, it is reasonable to start with the living cell, to study the effects of fixation on its visible constituents, to analyse their composition by cytochemical methods, and then to explore their fine structure by the use of the electron microscope. A manysided attack of this sort on a single kind of cell is much more likely to give valuable results than the independent study of different kinds of cells by different kinds of cytologists. I shall begin by giving an account of a particular piece of work just completed in the Departments of Zoology and Human Anatomy at Oxford by Mr. J. T. Y. Chou, Dr. K. F. A. Ross, and Dr. G. Meek. Although in this account I shall deal only with a single cytoplasmic inclusion in a single kind of cell, yet you will notice the general implications of my remarks and their bearing on problems of fixation.

CYTOCHEMISTRY AND ELECTRON MICROSCOPY The Preservation of Cellular Ultrastructure and Enzymatic Activity by Aldehyde Fixation

The aldehydes introduced in this paper and the more appropriate concentrations for their general use as fixatives are: 4 to 6.5 per cent glutaraldehyde, 4 per cent glyoxal, 12.5 per cent hydroxyadipaldehyde, 10 per cent crotonaldehyde, 5 per cent pyruvic aldehyde, 10 per cent acetaldehyde, and 5 per cent methacrolein. These were prepared as cacodylateor phosphate-buffered solutions (0.1 to 0.2 ...

Visualization of identified GFP-expressing cells by light and electron microscopy.

We have developed a procedure for visualizing GFP expression in fixed tissue after embedding in LR White. We find that GFP fluorescence survives fixation in 4% paraformaldehyde/0.1% glutaraldehyde and can be visualized directly by fluorescence microscopy in unstained, 1 microm sections of LR White-embedded material. The antigenicity of the GFP is retained in these preparations, so that GFP loca...

Hydroxypropyl Methacrylate, a New Water-miscible Embedding Medium for Electron Microscopy

Aldehyde-fixed rat tissues were variously dehydrated and impregnated in water-miscible 2-hydroxypropyl methacrylate (HPMA) containing 3 to 20 per cent water and 0.1 per cent alpha,alpha-azobisisobutyronitrile as catalyst for subsequent polymerization with ultraviolet light. Heat polymerization was also effective. Blocks of embedded tissue readily gave ultrathin sections, which required staining...
